Hemophilia B is a rare, inherited bleeding disorder caused by a deficiency in functional coagulation Factor IX (FIX). This deficiency impairs the blood's ability to clot, leading to prolonged and excessive bleeding after injuries, surgery, or even spontaneously. It is an X-linked recessive disorder, primarily affecting males.
Hemophilia B is a genetic disorder caused by mutations in the F9 gene located on the X chromosome. This gene provides instructions for making coagulation Factor IX. Most cases are inherited from a carrier mother or arise from spontaneous mutations.
Factor IX is a crucial component of the intrinsic pathway of the coagulation cascade. In hemophilia B, mutations in the F9 gene lead to either a reduced amount of FIX or the production of a dysfunctional FIX protein. When an injury occurs, the coagulation cascade is initiated, but the deficiency of functional FIX significantly compromises the formation of the tenase complex, which amplifies thrombin generation. This results in a poorly formed and unstable fibrin clot, leading to prolonged bleeding. The severity of hemophilia B correlates with the level of functional FIX in the blood.
Hemophilia B is estimated to affect approximately 1 in 25,000 to 30,000 live male births worldwide. It is less common than hemophilia A (Factor VIII deficiency). The prevalence is similar across all ethnic groups.
